Research in Dr. Meezan's laboratory is focused on the biochemical pharmacology of the alkylmaltosides in diabetes mellitus and cystic fibrosis. The alkylmaltosides are nontoxic, nonionic, mild surfactant agents which have been found by us to have significant pharmacological potential. They serve as acceptor substrates for glucose transfer by glycogenin and glycogen synthase, the initiating and elongating enzymes of glycogen synthesis respectively, and therefore, have the potential to modulate the synthesis of glycogen in diseases such as diabetes mellitus. These agents are also active in increasing the chloride permeability of epithelial cells with defective chloride channels in cystic fibrosis and thus, show promise as therapeutic agents in this disease. Finally, the alkylmaltosides are versatile absorption enhancing agents which allow the systemic effects of several hormones, including insulin and glucagon to be achieved via nasal and ocular administration of these polypeptides, thus, obviating the need for injections. All of these properties of the alkylmaltosides are currently under active investigation in our laboratory.
