Mouse Pulmonary Analysis Core Facility

  • Provides LHC members access to a state-of-the-art mechanical ventilator designed specifically to test lung function in mice.
  • Instruction in bronchioalveolar lavage and differential cell analyses of murine samples to LHC members.

Applications to pulmonary biospecimens include, but are not limited to, analyses of respiratory mechanics, immune cell trafficking, and inflammatory mediator identification.

The core facility has the following instrumentation and instruction:

  • The flexiVentTM system from SciReq. This system includes a computer and a novel software-controlled ventilator for mice designed to generate accurate and detailed measurements of respiratory mechanics. With this system, resistance and compliance measurements and pressure-volume loops can be obtained. The equipment also allows the investigator to conduct controlled aerosol delivery and to distinguish between central and peripheral lung mechanics.
  • MouseOx Small Animal Pulse Oximeter from Starrlife Sciences. This instrument measures arterial oxygen saturation, heart rate and breath rate all from onenon-invasive sensor. A direct application of this system is its ability to monitor the depth of anesthesia via measurements of heart rate.
  • Bronchoalveolar lavage (BAL) analysis. During this instruction, mice are anesthetized with ketamine / xylazine and then lavaged with 1 ml of 0.9% saline. Collected BAL samples are centrifuged to pellet the cellular fraction; the supernatant can be utilized for cytokine analysis.
  • Differential cell analysis. For this instruction, cell types are differentiated on cytospin preps using Diff-Quik stain. Cell differentials can then determined from at least 500 leukocytes using standard hematological criteria.
