PGP/Ac-PGP Method

  2. Acetylated-Proline-Glycine-Proline (Ac-PGP)

Chromatography

Liquid chromatography tandem mass spectrometry (LC-MS) was carried out with a Shimadzu Prominence HPLC system and SCIEX 6500 Qtrap mass spectrometer. An aliquot of 10 µl of each sample was injected onto an Jupiter 4 µm C18 150 x 2.0 mm column at 40oC for gradient separation. Mobile phases were: A, ddH2O with 0.1% Formic Acid; 0.5% DMSO; and B, Acetonitrile with 0.1% Formic Acid; 0.5% DMSO. Gradient schedule started at 5% B and was linearly increased to 100% B at 2.5 minute, 100% B hold to 2.75 minutes, 5% B at 3.0 minutes and held at 5% B until 5.0 minutes to re-equilibrate. Time per injection was 5.0 minutes.
