Atmospheric Pressure Ionization Liquid Chromatography tandem mass spectrometry (APCI-LC-MS) was carried out in the negative polarity with a Shimadzu Prominence HPLC system and SCIEX 4000 mass spectrometer. An aliquot of 20 µl of each sample was injected onto an Xtera C18, 5 µm 100 x 2.1 mm column for an  isocratic mobile phase separation that consisted of methanol and 0.05% aqueous ammonium hydroxide solution (98:2) at a flow rate of 1.0 mL/min. Total run time is 5 minutes.

Extraction Protocols:

Fifty µL of plasma was mixed with 200 µL of 1% (v/v) acetic acid in methanol and 50 µL of thymol (3000 nM) as per internal standard, then vortexed and centrifuged at 1,800 ×g for 10 min. Supernatant was directly analyzed.
