and consultation to users in the following
Research Capabilities useful In Structural Biology and Drug Design and Discovery Efforts: - Quality Control ln Synthetic Chemistry and Medicinal Chemistry Drug Discovery/Design Projects by 1D/2D-NMR (1H, 13C, 19F, 31P, 195Pt. etc).
- Determination of the three-dimensional structures of proteins and their complexes (protein-protein, protein-nucleic acid, protein-carbohydrate).
- Protein complexes with low molecular weight compounds (including low-affinity compounds). The bound·structure of the compound can be used in Structure-based design of improved lead compounds and ultimately drugs.
- STD-NMR Screening (a binding assay) of single Compound or Compound Libraries to identify "hits'' recognized by a target protein. Group epitope mapping of ligands. STD-NMR/CORCEMA-ST analyses of bound-ligand conformations.
- Fragment-based discovery & design (lead optimization) of new drugs by: (a) SAR-by-NMR (a technique pioneered by the Abbott Laboratories, widely used in Pharmaceutical Industry), and the (b) ILOE technique
- Membrane proteins in solution (proteins in micelles and bicelles).
- NMR of cells, cell extracts, and Metabonomlcs-by-NMR
Equipment
The Facility provides access to the following stale-of-the-art NMR instrumentation
1.850 MHz Bruker Avance III NMR Spectrometer (Ascend) with a CryoProbe (installation in progress)
2.700 MHz Bruker Avance II NMR with a CryoProbe
3.600 MHz Bruker Avance III NMR Spectrometer with a CryOProbe
4.500 MHz Bruker Avance NMR Spectrometer with TXI and TBI probes
