iPSC generation from primary human cells (3 clones) ($8000)
This service covers the generation of iPSCs from human PBMCs or other primary cell types (e.g., fibroblasts) provided by UAB investigators or obtained from commercial sources. Appropriate IRB approval may be required for human samples, when applicable. Primary cells will be reprogrammed using Sendai virus expressing Yamanaka factors, followed by isolation and expansion of iPSC clones. Three independent iPSC clones that pass quality control will be provided to the investigator.

PBMC isolation Support ($800/sample)
This service covers the isolation and cryopreservation of PBMCs from freshly collected human blood provided by UAB investigators for subsequent iPSC generation. Appropriate IRB approval may be required, when applicable.

iPSC clone expansion ($650/clone)
This service covers the expansion of investigator-provided iPSC clones. Expanded iPSCs will be provided to the investigator as a live culture or six cryopreserved vials.

iPSC storage and maintenance ($250/5 years)
This include the liquid nitrogen storage of up to 5 vials of each iPSC in 2 independent UAB locations

iPSC restocking fee ($150/vial)
The fees covers the expansion and restocking fee of requested iPSC lines.

iPSC AAVS1 Transgenic insertion (3 clones) ($10,250)
This service covers the generation of engineered iPSC lines where using a transgene is inserted into the safe harbor AAVS1 site using CRIPSR mediated targeting. Cost and labor involved in the generation of the targeting vector is on the PI, but TGEM is willing to assist in eth design and outsource for DNA synthesis at the PI expense. Some example of this include inducible Cre lines, fluorescent biosensors, or reporters of differentiation.

iPSC engineered KO (3 clones) ($8000)
This service covers the generation of CRIPSR mediated indels in iPSC that result in bi-allelic frame shifts in an early exon (or desired exon) such as to generate a null allele. All reagents are covered in this cost. 3 independent sequence verified clones will be provided.

iPSC correction to WT (3 clones) ($11,000)
This service covers the conversion of one patient mutation in an iPSC line to a wildtype sequence using CRISPR technology. All reagents are covered in this cost. 3 independent sequence verified clones will be provided. Note conversion of compound het mutation will require two independent services to convert to complete wildtype sequence.

iPSC engineered patient mutation (3 clones) ($11,000)
This service covers the conversion of a “wildtype” iPSC line to mutation containing iPSC line using CRISPR technology. This service is most often used to generate an isogenic series of iPSC with different patient mutation to compare the impact across the same genetic background. All reagents are covered in this cost. 3 independent sequence verified clones will be provided.
Service Guarantee: For TGEM IPSC services we will only bill upon providing final product of service. If reagent can not be generated no fee will be incurred.
